Growth Hormone Secretagogues

Gonadorelin

Also known as: GnRH

Gonadorelin is a synthetic decapeptide identical to endogenous gonadotropin-releasing hormone (GnRH/LHRH; sequence pyroGlu-His-Trp-Ser-Tyr-Gly-Leu-Arg-Pro-Gly-NH2). It was historically FDA-approved as Factrel (diagnostic GnRH stimulation test) and Lutrepulse (pulsatile IV pump for hypothalamic amenorrhea); both products are discontinued in the US. Gonadorelin remains on the FDA Category 1 bulk substances list and is widely compounded, most commonly as an off-label adjunct to TRT intended to preserve HPG-axis signaling.

Diagnostic Usefulness of Subcutaneous Triptorelin Stimulation Testing in the Evaluation of Central Precocious Puberty in Girls

The limited availability of gonadorelin has resulted in an increased use of subcutaneous triptorelin to diagnose central precocious puberty; however, studies validating its diagnostic value remain limited.

Diagnostic performance of a simplified GnRH stimulation test in central precocious puberty: a multicentre study

Abstract Background Diagnosis of central precocious puberty (CPP) relies on biochemical confirmation of premature activation of the hypothalamic–pituitary–gonadal axis, with the GnRH stimulation test considered the reference standard.

Increasing gonadorelin hydrochloride dose at the first GnRH of breeding Ovsynch enhances luteinizing hormone release, ovulatory response, and pregnancy in first-service lactating dairy cows

The objective of this study was to evaluate the effect of administering 200 µg versus 100 µg of GnRH as gonadorelin hydrochloride (Factrel; Zoetis, Parsippany, NJ) at the first GnRH injection (G1) of the breeding Ovsynch in a Double-Ovsynch (DO) protocol on pregnancy per artificial insemination (P/AI) at first service…

Estradiol treatment and double dose of GnRH for resynchronization protocols and timed artificial insemination in lactating dairy cows

The aim of this study was to compare pregnancy per AI (P/AI) in resynchronization (Resynch) protocols using different hormonal combinations (estradiol benzoate [EB], a double dose of GnRH, or EB with a double dose of GnRH) administered on the day of progesterone (P4) device insertion.

Ovarian remnant syndrome in a 2-year-old cat with inconclusive diagnostics

Case summary A 2.5-year-old, spayed female domestic shorthair cat was evaluated for estrus-like behavior (lordosis, vocalization and rolling) occurring on a cyclical basis every 3 weeks. Clinical signs began 6 months after ovariohysterectomy and exploratory abdominal surgery was performed; no ovarian tissue was found.

Stress-related cortisol and progesterone concentrations influence the success of ovulation induction in estrous cats

Queens are induced ovulators, and ovulatory success varies with hormonal status and stress.
